A biomimetic assay to quantify the interaction kinetics of synaptic complexes

Adhesion is a fundamental mechanism regulating many biological processes. At the molecular level, adhesion is mediated by ligand-receptor pairs expressed by all the cell types of the organism. At the synaptic level, adhesion is mediated by a wide-range of partners such as the Neurexin and Neuroligin pair. However, little is known about the stoichiometry and dynamics of these molecules. Studying these paramaters in vivo proved to be highly challenging and so there is a strong need for a simplified model. This is the reason why we have developed, in collaboration with Olivier Thoumine (IINS, Bordeaux, France), a biomimetic system based on the immobilization of Fc-tagged ligands on micropatterned substrates, combined with live-cell imaging (SPT & FRAP) of fluorescently tagged receptors in heterologous cells to extract biophysical parameters. Using mathematical simulations (Lagardère et al., 2020), we aim at understanding the dynamic of ligand-receptor pairs and thus better understand its involvement in disease progression.
